Isolation and characterization of polyhydroxyalkanoate-producing bacteria from seawater samples (Tumaco)

Abstract. Introduction. Polyhydroxyalkanoate (PHA) is a useful, carbon-neutral polymer that can be produced from renewable carbon sources by microorganisms, making it sustainable and environmentally friendly material. Currently, production of the biopolymer not competitive in terms cost yield; however, different bacterial strains may provide economic viability, especially since substituting biodegradable plastics for nondegradable synthetic has drawn interest both academia commercial world. Objective. The aim this work was to describe isolation partial characterization PHA-producing bacteria isolated five sites (gas stations) coastal regions Tumaco Island, Nariño-Colombia, classify physicochemical parameters granules show biopolymer. Materials methods. A viable colony staining method using Nile red used preliminarily screen bacteria. Colonies were isolated, characterized via biochemical, microbial, molecular methods, tested antimicrobial susceptibility fermentation. Results. More than thirty-eight identified as potential PHA-positive isolates screening approach but, just one PHA (T2-25A). All resistant metronidazole, ampicillin, trimethoprim sulfamethoxazole, cephalothin, ceftriaxone, cefazolin, 27.3 % novobiocin. Conclusions. One promising isolate obtained. Nevertheless, information will complement future studies conditions necessary produce PHA.
 Moreover, antibiotic resistance data have attracted attention, because origin source waters isolates.
